      you can use Amazon add a product tool to check if the UPC is new or have been used, open add a product tool and put UPC as product identifier, if it is new no error will be show and if it has been used Amazon will give error.

      There are different websites which allow you to check the validation of UPCs and you can also check it from the amazon, if you have seller central account, you can go to add to product, enter the UPC if you dont get any error then the UPC is ok.
